Definition:
The process of selective retardation of one or more components of a fluid solution as the fluid uniformly percolates through a column of finely divided substance, or through capillary passageways. The retardation results from the distribution of the components of the mixture between one or more thin phases and the bulk fluid, as this fluid moves countercurrent to the thin, stationary phases. The process is used for analysis and separation of mixtures of two or more substances. Among the many variations of the process are gas chromatography (the specimen in gaseous form is passed through a capillary tube line with a liquid or solid phase), paper chromatography (a drop of the specimen is placed near one end of a porous paper), ion exchange chromatography, thin layer chromatography (the sample is placed on an absorbent cake spread on a smooth glass plate), gel permeation chromatography and reverse phase liquid chromatography (RPLC).
